Is total serum protein a good indicator for welfare in reared sea bass (Dicentrarchus labrax)? ArchiMer
Coeurdacier, Jean-luc; Dutto, Gilbert; Gasset, Eric; Blancheton, Jean-paul.
Reared sea bass were submitted to six stressful situations: hyperoxia with or without hypercapnia, increased stocking density in an open flow or recirculating system, transfer to another tank and nodavirus injection. The potentially negative impact of these factors on the lives of sea bass was investigated by measuring 9 water parameters and 19 fish parameters including total serum protein (TSP). TSP has already been used to evaluate stress. The present study investigates data of previous and new experiments, concentrating on the potential use of TSP as a routine indicator to assess welfare in sea bass reared on fish farms. In the current experiment, oxygen and carbon dioxide were seen to affect levels of TSP, but alterations were too erratic to enable...

Le concept de biosécurité en pisciculture marine : prévention de l'encéphalite virale chez l'ombrine subtropicale (Sciaenops ocellatus) en Martinique ArchiMer
Adee, Fabrice.
In 1985, red drum (Sciaenops ocellatus) was introduced in Martinique in arder to replace sea bass fish farming. Nodavirus antibodies were detected in the broodstock at the IFREMER station in Robert. Nodavirus is responsible for viral nervous necrosis, a disease that is spreading quickly in worlwide marine aquaculture. A biosecurity process was initiated at the Robert hatchery to select seronegative breeders and produce nodavirus free larvae. This process, based on a serological follow-up of fish antibodies by ELISA, allowed a reduction of the number of seropositive fish. A complementary serological study revealed a low incidence of nodavirus contamination in nodavirus free larvae placed in sea cages on different production sites

Le système immunitaire du bar Dicentrarchus labrax. Immunoglobulines, cellules productrices d'immunoglobulines, ontogenèse applications à une pathologie virale néonatale ArchiMer
Breuil, Gilles.
The sea bass, Dicentrarchus labrax, has been subjected to intensive aquaculture on the Mediterranean cost for ten years. Marine fish hatcheries are key factors in the aquaculture field, and face different pathologies, especially one of viral origin (nodavirus) that could challenge their feasibility. Consequently, we have tried to provide some data on larvae and spawning stocks. Firstly, we have studied the immunoglobulins, components of the specific humoral immunity, and their role as antibodies. On adult basses, these immunoglobulins are tetrameric molecules with high molecular weight (850 kD), made of heavy (H) - 80 kD and light (L) - 26 kD chains. In the eggs, simplest forms such as monomeric or dimeric are brought to the fore; they are located under...

Quantitative relationship of two viruses (MrNV and XSV) in white-tail disease of Macrobrachium rosenbergii ArchiMer
Zhang, Hj; Wang, Hm; Yuan, Jf; Li, Lj; Zhang, Jh; Bonami, Jean-robert; Shi, Zl.
Macrobrachium rosenbergii nodavirus (MrNV) and extra small virus (XSV) were purified from diseased freshwater prawns M rosenbergii and used to infect healthy post-larvae (PL) by an immersion method. Three groups of prawns were challenged with various combined doses of MrNV and XSV. Signs of white-tail disease (WTD) were observed in Groups 1 and 2, which had been challenged with combinations containing relatively high proportions of MrNV and low proportions of XSV. By contrast there was little sign of WTD in Group 3, which had been challenged with a higher proportion of XSV than MrNV. A 2-step Taqman real-time RT-PCR was developed and applied to quantify viral copy numbers in each challenged PL. Results showed that genomic copies of both viruses were much...

Sea bass Dicentrarchus labrax nervous necrosis virus isolates with distinct pathogenicity to sea bass larvae ArchiMer
Breuil, Gilles; Mouchel, Olivier; Fauvel, Christian; Pepin, Jean-francois.
Reproduction of nodavirus disease was performed by experimental infection of sea bass eggs during fertilization or at larval stage 4 with 2 genetically distinguishable nodavirus strains (Sb1 and Sb2) isolated from sea bass collected along the Atlantic and Mediterranean French coast. The pathogenicity of the virus strains was assigned after detection of the virus by ELISA and immunohistochemistry (IHC). The Atlantic (Sb1) strain was more pathogenic than the Mediterranean (Sb2) strain during the fertilization step whilst both strains were pathogenic following experimental exposure of 4 d old larvae. Virus lesions developed in the brain 4 to 6 d following experimental exposure. Experimental ELISA proved very sensitive for detecting the nodavirus in Sb1 or Sb2...
